ALN-6222 is an investigational, subcutaneously administered RNA interference (RNAi) therapeutic designed to treat obesity by targeting the *INHBE* (Inhibin subunit beta E) gene. Developed by Alnylam Pharmaceuticals, the drug utilizes a small interfering RNA (siRNA) conjugated to N-acetylgalactosamine (GalNAc) to specifically target hepatocytes in the liver. The *INHBE* gene encodes a hepatokine that has been identified through human genetics as a regulator of adipose tissue distribution and metabolic health. Genetic evidence suggests that silencing or loss of *INHBE* is associated with reduced abdominal fat and a lower risk of metabolic diseases such as type 2 diabetes. ALN-6222 aims to replicate this protective genetic effect to induce weight loss and improve metabolic parameters.
