ALN-AGT02 is a second-generation RNA interference (RNAi) therapeutic being developed by Alnylam Pharmaceuticals in collaboration with Regeneron Pharmaceuticals for the treatment of hypertension. It consists of a small interfering RNA (siRNA) designed to specifically target and silence the messenger RNA (mRNA) for angiotensinogen (AGT), a liver-produced precursor protein that is the sole source of all angiotensin peptides in the renin-angiotensin-aldosterone system (RAAS). By reducing AGT production at its source, ALN-AGT02 aims to provide a potent and durable reduction in blood pressure with infrequent dosing. This program is distinct from Alnylam's first-generation AGT-targeting siRNA, zilebesiran (ALN-AGT), and is currently in early clinical development.
