ALN-IL4R (also known as REGN6191) is an investigational small interfering RNA (siRNA) therapeutic designed to silence the expression of the interleukin-4 receptor alpha (IL-4Rα) subunit. Developed through a strategic collaboration between Alnylam Pharmaceuticals and Regeneron Pharmaceuticals, the drug utilizes RNA interference (RNAi) to degrade the mRNA encoding IL-4Rα. Because IL-4Rα is a shared component of both the IL-4 and IL-13 receptor complexes, its knockdown effectively inhibits signaling of both cytokines, which are central drivers of Type 2 (Th2) inflammation. This mechanism is biologically analogous to the monoclonal antibody dupilumab, but by acting at the pre-translational level, ALN-IL4R may offer advantages such as localized delivery to the lungs via inhalation for respiratory conditions and a potentially longer duration of action, allowing for infrequent dosing schedules.
