ALN-KHK is an investigational, subcutaneously administered RNA interference (RNAi) therapeutic developed by Alnylam Pharmaceuticals. It is designed to target and silence the expression of hepatic ketohexokinase (KHK), also known as fructokinase, the first enzyme in the pathway of fructose metabolism. By inhibiting KHK, ALN-KHK aims to reduce fatty acid synthesis and insulin resistance, potentially improving glycemic control in individuals with type 2 diabetes mellitus (T2DM). The drug utilizes N-acetylgalactosamine (GalNAc)-conjugated technology for targeted delivery to the liver. Preclinical studies have shown potent and durable reduction in KHK protein levels following administration. Clinical development has focused on overweight or obese adults with T2DM.
