Alnodesertib is a first-in-class, orally available, selective small molecule inhibitor of ataxia-telangiectasia and Rad3-related protein (ATR) and ataxia telangiectasia mutated (ATM) kinase, developed as an anti-cancer agent primarily for the treatment of solid tumors. By inhibiting ATR and ATM (DNA damage response kinases), alnodesertib interferes with tumor cell DNA repair mechanisms, particularly in tumors with defective DNA repair pathways, thereby leading to cancer cell death. It is designed to target tumors deficient in DNA repair genes, such as those with ATM, BRCA, or other homologous recombination deficiency alterations[1][3][5][7][9].
