Alocresti is a liposomal formulation of vinorelbine, a semi-synthetic vinca alkaloid with antineoplastic activity. Developed using proprietary liposomal encapsulation technology (Optisome) originally by Inex Pharmaceuticals, Alocresti was designed to improve the therapeutic index of vinorelbine by prolonging its circulation time and enhancing its accumulation within tumor tissues via the enhanced permeability and retention (EPR) effect. Mechanistically, vinorelbine binds to tubulin, inhibiting microtubule polymerization and disrupting mitotic spindle assembly, which results in cell cycle arrest and apoptosis. The drug was primarily developed by Talon Therapeutics and Spectrum Pharmaceuticals for the treatment of advanced solid tumors, including non-small cell lung cancer (NSCLC). Although some historical contexts list it alongside approved products, it did not receive FDA approval and its development was eventually discontinued by Spectrum Pharmaceuticals.
