Overview

Alosetron is a potent and selective small molecule antagonist of the serotonin 5-HT3 receptor. It is used for the management of severe diarrhea-predominant irritable bowel syndrome (IBS-D) in women who have not responded to conventional therapy. By blocking 5-HT3 receptors in the gastrointestinal tract, alosetron modulates serotonin-sensitive GI processes, reducing visceral pain, colonic transit, and GI secretions associated with IBS-D. The drug was originally withdrawn from the US market due to reports of serious adverse effects such as ischemic colitis but was later reintroduced under a restricted marketing program[1][2][3][4][8].
