Overview

Alpelisib + carboplatin is a combination therapy being investigated for the treatment of advanced solid tumors and human papillomavirus (HPV)-positive squamous cell carcinoma. Alpelisib (BYL719) is a small molecule inhibitor that selectively targets the p110α catalytic subunit of phosphatidylinositol 3-kinase (PI3Kα), a pathway frequently mutated or overactivated in various cancers, including those associated with HPV. Carboplatin is a platinum-based chemotherapeutic agent that induces DNA cross-links, interfering with DNA replication and transcription to trigger apoptosis. This combination is being evaluated in a Phase I/II clinical trial led by Dr. Pamela Munster at the University of California, San Francisco, to determine the safety, maximum tolerated dose, and efficacy in patients with HPV-positive malignancies.
