ALPHA-003 is a preclinical-stage small molecule drug candidate developed by Filamon Limited for the treatment of neurodegenerative diseases associated with abnormal protein aggregation, specifically tauopathies. The drug utilizes a novel anti-inflammatory approach by binding to structural proteins, including microtubule-associated protein tau, neurofilament light polypeptide (NfL), and TAR DNA-binding protein 43 (TDP-43), to protect them from inflammatory disruption. It specifically blocks the activating effects of the inflammatory ligand human Group IIA secretory phospholipase A2 (hGIIA) on these proteins, thereby preserving the integrity of neuronal microtubules and preventing brain cell death. Developed using Australian-designed deep-learning computational drug design technology, ALPHA-003 is orally bioavailable and capable of crossing the mammalian blood-brain barrier. It is currently undergoing preclinical testing with plans to enter clinical trials in 2026.
