alpha 2 integrin shRNA is a gene-silencing tool designed to reduce the expression of the alpha 2 integrin subunit (ITGA2). Integrin alpha 2 dimerizes with the beta 1 subunit to form the α2β1 heterodimer, which serves as a major cell surface receptor for collagen and plays a critical role in cell adhesion, migration, and tumor progression. In preclinical research, specifically in pancreatic cancer models, lentiviral-based delivery of this shRNA has been shown to significantly inhibit cancer cell adhesion and proliferation on collagen types I and IV. Although it demonstrates potent knockdown of ITGA2 mRNA and protein levels (often exceeding 90%), studies indicate that targeting the alpha 2 subunit may be less effective at inhibiting in vivo tumor growth and metastasis compared to targeting the beta 1 subunit, which appears to play a more central role in pancreatic cancer malignancy.
