Alpha-201 + interleukin-12 + interleukin-15 (also known as AMG 199) is an investigational oncolytic immunotherapy developed by Amgen. It consists of a second-generation, modified herpes simplex virus type 1 (HSV-1) vector engineered to selectively replicate within and lyse tumor cells. Beyond its direct oncolytic activity, the virus is designed to deliver and express two potent proinflammatory cytokines, interleukin-12 (IL-12) and interleukin-15 (IL-15), directly within the tumor microenvironment. IL-12 promotes a Th1 immune response and activates cytotoxic T cells and natural killer (NK) cells, while IL-15 supports the survival and proliferation of these effector cells. By combining local viral-induced cell death with the targeted expression of these cytokines, Alpha-201 aims to stimulate a systemic, durable anti-tumor immune response, potentially overcoming resistance to existing immunotherapies in patients with advanced solid tumors.
