This investigational cancer immunotherapy consists of alpha-type 1 polarized dendritic cells (αDC1) loaded with synthetic peptides for four glioma-associated antigen (GAA) epitopes: EphA2, IL-13Rα2, YKL-40, and gp100. Developed at the University of Pittsburgh by Dr. Frank Lieberman and Dr. Hideho Okada, the vaccine is administered via ultrasound-guided intranodal injection in combination with poly-ICLC (Hiltonol), a synthetic double-stranded RNA that acts as a Toll-like receptor 3 (TLR3) agonist. This combination is designed to induce a robust, tumor-specific CD8+ T-cell response in HLA-A2 positive patients with recurrent malignant gliomas. The treatment protocol includes an initial vaccination series followed by booster phases to maintain the immune response.
