Overview

Alpha DEC205-EBNA1 fusion antibody (αDEC-E1) is a research-stage biologic fusion protein vaccine candidate designed for the treatment and prevention of Epstein-Barr virus (EBV)-associated malignancies and symptomatic primary EBV infections, such as infectious mononucleosis. Developed by researchers at the University of Zurich and the University of Oxford in collaboration with Miltenyi Biotec, the biologic consists of a monoclonal antibody targeting the DEC205 receptor (clone MG38.2) fused to the EBV nuclear antigen 1 (EBNA1, amino acids 400-641). DEC205 is a type I cell surface receptor expressed primarily on conventional type I dendritic cells (cDC1s). By delivering the EBNA1 antigen directly to these dendritic cells, the fusion protein facilitates antigen cross-presentation to elicit robust CD4+ and CD8+ T-cell immune responses. Preclinical research has demonstrated that αDEC-E1 is particularly effective in heterologous prime-boost vaccination regimens, where it is used as a priming agent followed by a boost with viral vectors (such as adenovirus or MVA) encoding EBNA1, to establish immune control over EBNA1-expressing lymphomas.
