Alpha-fetoprotein peptide + glypican-3 peptide is a combination cancer vaccine composed of specific peptide fragments derived from two oncofetal antigens: alpha-fetoprotein (AFP) and glypican-3 (GPC3). These antigens are significantly overexpressed in hepatocellular carcinoma (HCC) but are largely absent in healthy adult tissues, making them ideal targets for immunotherapy. The vaccine works by stimulating the patient's immune system to generate antigen-specific CD8+ cytotoxic T lymphocytes (CTLs). These CTLs recognize the HLA-restricted peptides presented on the surface of tumor cells and induce apoptosis. This combination approach is designed to overcome tumor heterogeneity and reduce the risk of immune escape by targeting multiple markers associated with liver cancer cells. Most clinical development has been centered in Japan, often investigating the vaccine's ability to prevent post-operative recurrence or treat advanced HCC.
