Alpha-fetoprotein small interfering RNA (AFP-siRNA) is an RNA interference (RNAi) therapeutic designed to specifically target and silence the expression of the alpha-fetoprotein (AFP) gene. AFP is a glycoprotein normally produced during fetal development but is frequently overexpressed in hepatocellular carcinoma (HCC), where it serves as a diagnostic biomarker and a driver of tumor progression. By delivering siRNA sequences complementary to AFP mRNA—often encapsulated in delivery vehicles like polylactic-co-glycolic acid (PLGA) nanoparticles—the drug triggers the catalytic degradation of the target mRNA, thereby reducing AFP protein levels. Research indicates that AFP-siRNA can inhibit tumor cell proliferation and may work synergistically with multi-kinase inhibitors such as sorafenib or sunitinib to enhance anti-tumor activity in liver cancer models.
