Overview

Alpha-fucosylceramide (specifically α-L-fucosylceramide) is a synthetic glycolipid and a structural analogue of alpha-galactosylceramide (α-GalCer). It consists of an α-L-fucose sugar moiety linked to a ceramide backbone. The compound acts as a ligand for the CD1d receptor on antigen-presenting cells, which in turn activates invariant natural killer T (iNKT) cells. This activation triggers the production of various cytokines, including both Th1 and Th2 types. Research has explored its potential in treating cancer, autoimmune diseases like multiple sclerosis and arthritis, and infectious diseases. However, some studies, such as those presented at AACR 2010, indicated that α-fucosylceramide lacked significant anti-tumor activity in certain models compared to other glycosylceramides. It is currently a research-stage compound with no known commercial developer or clinical trials.
