Overview

The alpha-gal positive pancreatic ductal adenocarcinoma tumor lysate vaccine is an experimental autologous cancer immunotherapy designed to treat pancreatic ductal adenocarcinoma (PDAC). The vaccine is produced by taking resected tumor tissue from a patient, creating a cell lysate, and enzymatically processing it to express alpha-galactosyl (α-gal) epitopes on surface glycoproteins. Humans naturally possess high titers of anti-Gal antibodies; when the vaccine is administered, these antibodies bind to the α-gal epitopes, leading to the opsonization of the tumor antigens. This process facilitates the uptake of tumor-associated antigens by antigen-presenting cells (APCs), such as dendritic cells, which subsequently prime and activate tumor-specific T cell responses. This approach aims to overcome the poor immunogenicity of PDAC by leveraging the innate immune system to recognize a broad spectrum of tumor antigens, including those found on pancreatic cancer stem cells.
