The alpha-gal tumor lysate vaccine is an experimental immunotherapy designed to target cancer stem cells (CSCs) and differentiated tumor cells. The vaccine is produced by engineering tumor cells (such as the PANC1 pancreatic cancer line) to express alpha-gal epitopes (Galα1-3Galβ1-4GlcNAc-R) through transfection with the alpha-1,3-galactosyltransferase (α1,3GT) gene. Because humans naturally possess high titers of anti-Gal antibodies, the presence of alpha-gal epitopes on the tumor lysate facilitates the formation of immune complexes. These complexes are efficiently opsonized and internalized by antigen-presenting cells, leading to the activation of tumor-specific T cells. Research indicates this approach can effectively target CSC markers like CD44 and CD24, potentially preventing tumor regrowth and metastasis.
