Alpha-galactosylceramide is a synthetic glycolipid originally derived as an analogue of agelasphin-9b from marine sponges. It acts as a potent immunostimulant by activating natural killer T (NKT) cells through presentation by CD1d molecules on antigen-presenting cells, leading to the activation and expansion of NKT cells. This activation results in downstream immune effects, including dendritic cell maturation and B-cell activation via IL-4 production. Alpha-galactosylceramide has demonstrated antitumor activity in animal models and has been investigated as an adjuvant for vaccines and for its potential to induce T-cell immunity against cancer and viral infections[1][2][3][5][6]. Development efforts have included use in hepatitis B, hepatitis C, lung cancer, and as a mucosal vaccine adjuvant.
