Alpha-hydroxy glycineamide (αHGA) is a small molecule antiviral agent and the active metabolite of the synthetic tripeptide glycyl-prolyl-glycine-amide (GPG-NH2). It exhibits potent antiviral activity against HIV-1 by interfering with viral capsid formation and targeting the envelope precursor protein gp160 for degradation via the ER-associated protein degradation (ERAD) pathway. This results in reduced incorporation of envelope proteins into progeny virions, rendering them fusion-incompetent. The compound enters cells primarily through passive diffusion and acts independently of serum type. Its mechanism is novel among antivirals, as it specifically disrupts HIV-1 Env maturation rather than directly inhibiting viral enzymes or entry[1][2][4].
