Overview

Alpha-linolenic acid (ALA) is an essential omega-3 polyunsaturated fatty acid found in plant sources like flaxseed, walnuts, and chia seeds. In the context of neurodegenerative research, ALA serves as a metabolic precursor for the endogenous synthesis of long-chain omega-3 fatty acids, specifically docosahexaenoic acid (DHA). This metabolic pathway is particularly relevant for APOE4 carriers, who often exhibit impaired DHA transport across the blood-brain barrier (BBB) and reduced brain DHA levels. By providing ALA, researchers aim to bypass transport limitations and support cerebrovascular health, BBB integrity, and cognitive function in populations at high risk for Alzheimer's disease, such as those with mild cognitive impairment (MCI). ALA also acts as a signaling molecule, activating G protein-coupled receptors and peroxisome proliferator-activated receptors (PPARs) to exert anti-inflammatory and neuroprotective effects.
