Alpha-mannosylceramide (α-ManCer) is a synthetic glycolipid and monoglycosyl ceramide characterized by an alpha-linked mannose sugar moiety. It functions as a potent agonist for the Vα19-Jα26 invariant natural killer T (NKT) cell repertoire, a unique subset of T cells that recognize glycolipid antigens presented by the non-classical MHC-like molecule CD1d. While structurally related to the prototypical NKT agonist alpha-galactosylceramide (α-GalCer), α-ManCer has shown distinct biological activities in preclinical models. For instance, research has indicated that α-ManCer does not induce significant anti-tumor immunity in certain murine models of colon carcinoma and melanoma, unlike its beta-linked counterpart (β-ManCer). It is primarily utilized as a research tool in immunology to investigate NKT cell activation and potential immunotherapeutic strategies.
