Drug intelligence / Profile preview
An alpha-synuclein radiotracer is a radiolabeled imaging agent designed to selectively bind to pathological aggregates of alpha-synuclein (α-syn) in the brain. These tracers are utilized in positron emission tomography (PET) or single-photon emission computed tomography (SPECT) imaging to non-invasively visualize and monitor the progression of synucleinopathies, such as Parkinson's disease (PD), multiple system atrophy (MSA), and dementia with Lewy bodies (DLB). Developing these tracers has been historically challenging due to the low abundance of α-syn aggregates compared to amyloid-beta or tau, and the requirement for high selectivity over other misfolded proteins. Several candidates, including [18F]ACI-12589 (developed by AC Immune), [11C]MODAG-005, and [18F]MODAG-009 (developed by MODAG), are currently undergoing clinical evaluation.
