Drug intelligence / Profile preview
Alpha-synuclein shRNA is a gene-silencing therapeutic platform designed to downregulate the expression of alpha-synuclein (aSyn), a protein whose pathological aggregation into Lewy bodies is a hallmark of Parkinson's disease and other synucleinopathies. By utilizing short hairpin RNA (shRNA) delivered via viral vectors (such as adeno-associated virus or lentivirus) or non-viral vehicles (such as exosomes or nanoparticles), this approach targets the SNCA transcript to reduce endogenous monomeric alpha-synuclein levels. Lowering the pool of available alpha-synuclein monomers has been shown to mitigate pathological seeding, reduce aggregation, and protect against neurodegeneration in preclinical models of Parkinson's disease.
