Overview

Alpha-tocopheryloxyacetic acid (α-TEA) is a semi-synthetic, orally bioavailable ether derivative of alpha-tocopherol (vitamin E). It is a small molecule drug developed as an antineoplastic and immunostimulatory agent. α-TEA selectively induces apoptosis and autophagy in tumor cells, leading to direct cytotoxic effects. The autophagosomes generated carry tumor-associated antigens, enhancing cross-presentation by antigen-presenting cells and stimulating a T cell-mediated Th1 immune response. This results in increased cytotoxic T lymphocyte activity against cancer cells and reduced regulatory T cell differentiation within the tumor microenvironment. α-TEA also modulates cytokine release, increasing IFN-gamma while decreasing IL-4 levels, further promoting anti-tumor immunity. Preclinical studies have shown efficacy in various cancer models including breast cancer, with ongoing phase 1 clinical trials for several malignancies[1][2][6][10].
