The alpha-type-1-polarized dendritic cell vaccine (αDC1) is an autologous cell-based cancer immunotherapy developed by Dr. Pawel Kalinski and colleagues. This vaccine consists of dendritic cells generated from a patient's own peripheral blood mononuclear cells and matured using a specialized cytokine cocktail containing IFN-alpha, polyinosinic:polycytidylic acid (poly-I:C), TNF-alpha, IL-1beta, and IFN-gamma. The αDC1 cells are characterized by three key features: a fully mature status, responsiveness to secondary lymphoid organ chemokines (CCR7 ligands), and high interleukin-12p70 (IL-12p70) production capacity. These dendritic cells are loaded with tumor-associated antigens, such as apoptotic autologous tumor cells, and administered to patients to induce potent cytotoxic T lymphocyte (CTL) responses. The vaccine aims to stimulate the patient's immune system to recognize and destroy tumor cells by promoting Type-1 immunity and enhancing CTL infiltration into tumor tissues.
