**Alpha1-oleate** is a novel protein-lipid complex consisting of the 39-residue alpha-helical N-terminal peptide of human alpha-lactalbumin bound to multiple molecules of oleic acid. It originates from research on the HAMLET complex (Human Alpha Lactalbumin Made Lethal to Tumor cells) and reproduces its tumoricidal effects. The mechanism of action includes direct induction of apoptosis-like cell death in tumor cells (primarily bladder cancer), inhibition of oncogene and cancer-related gene networks, significant disruption of tumor microenvironment pathways (including angiogenesis, tumor invasion, and metastasis), and modulation of immune responses—specifically, broad anti-inflammatory and immune-inhibitory effects. Alpha1-oleate’s action is conformation-dependent, utilizing structural flexibility to expose tumor-selective motifs that accelerate tumor cell death. It is administered intravesically and has demonstrated strong, dose-dependent anti-tumor efficacy in clinical trials, especially for non-muscle invasive bladder cancer (NMIBC), with minimal toxicity limited to local instillation site irritation[2][3][5].
