**alphaASGR1-RSPO2-RA** is an experimental hepatocyte-targeted R-spondin mimetic protein designed to selectively enhance Wnt signaling in the liver. It consists of an anti-ASGR1 antibody fused to a mutant RSPO2 domain engineered to eliminate LGR4-6 binding while preserving binding to the Wnt negative regulators ZNRF3 and RNF43, thereby promoting stabilization of Wnt receptors and amplifying endogenous Wnt pathway activity specifically in ASGR1-expressing hepatocytes. Preclinical studies showed liver-selective induction of Wnt target genes, hepatocyte proliferation, and improved liver function in mouse models of liver injury, positioning it as a regenerative biologic concept for liver diseases. The molecule appears to be a research-stage predecessor or prototype related to later Surrozen hepatocyte-targeted RSPO mimetics such as SZN-043.
