Overview

AlphaPARP is a targeted alpha-particle radioligand therapy (TAT) being developed by Trevarx Biomedical for the treatment of various solid tumors. The therapeutic consists of a small molecule ligand that selectively binds to Poly (ADP-ribose) polymerase 1 (PARP1), conjugated to the alpha-emitting radioisotope Astatine-211 (211At). PARP1 is an enzyme involved in DNA repair that is frequently overexpressed in cancers such as ovarian, prostate, and breast cancer. By delivering high-LET (linear energy transfer) alpha radiation directly to the site of PARP1 expression, AlphaPARP induces complex, difficult-to-repair double-strand DNA breaks in cancer cells while sparing adjacent healthy tissue due to the short range of alpha particles. The program is currently in IND-enabling studies for relapsed/refractory ovarian cancer and preclinical development for other indications including metastatic castration-resistant prostate cancer (mCRPC) and triple-negative breast cancer (TNBC).
