The alphaTIGIT-guided IL-15 mimetic is an engineered bispecific antibody-based cytokine agonist designed to overcome the systemic toxicity and structural instability associated with native IL-15. Developed using AlphaFold3-assisted structural modeling, this molecule combines an IL-15 mimetic with a TIGIT-targeting domain. This 'cis-targeting' strategy ensures that the cytokine activity is precisely directed toward TIGIT-expressing immune cells, specifically activating and expanding intratumoral CD8+ stem-like T cells. In preclinical models, this approach has demonstrated potent antitumor activity and a favorable safety profile compared to non-targeted IL-15 mimetics, which often show limited in vivo efficacy. The construct aims to bridge the gap between immune checkpoint blockade and cytokine therapy by providing targeted immune stimulation within the tumor microenvironment.
