ALPK1-IN2 (also known as compound T001) is a potent small molecule inhibitor of alpha-kinase 1 (ALPK1), an atypical protein kinase involved in innate immune responses and inflammatory signaling. It exhibits an IC50 of 95 nM against ALPK1 and also inhibits NF-κB signaling with an IC50 of 1.31 μM. By inhibiting ALPK1 kinase activity, the compound disrupts the downstream TIFA/TRAF6/NF-κB signaling cascade. Research suggests that ALPK1 inhibition can decrease the proliferation of diffuse large B-cell lymphoma (DLBCL) cells and increase apoptosis, potentially acting synergistically with Bcl-2 inhibitors like venetoclax to overcome drug resistance. While it is being investigated for potential therapeutic applications in autoinflammatory diseases, metabolic disorders, and certain cancers, ALPK1-IN2 is currently categorized as a research tool compound and is not approved for human or veterinary use.
