Drug intelligence / Profile preview
Alrizomadlin + lisaftoclax (APG-115 + APG-2575) is an investigational combination regimen consisting of two orally active small-molecule apoptosis-targeted agents developed by Ascentage Pharma. Alrizomadlin is a highly selective inhibitor of the MDM2-p53 protein-protein interaction, which restores the tumor suppressor activity of p53 and induces apoptosis in TP53 wild-type cancer cells. Lisaftoclax is a selective Bcl-2 inhibitor that triggers mitochondrial outer-membrane permeabilization and caspase activation to promote apoptosis. By simultaneously targeting the MDM2-p53 and Bcl-2 apoptotic pathways, the combination aims to achieve synergistic antitumor activity and overcome resistance to single-agent Bcl-2 inhibitors like venetoclax. The combination is currently undergoing Phase Ib/II clinical evaluation for the treatment of pediatric solid tumors, including rhabdomyosarcoma, neuroblastoma, and other soft-tissue sarcomas, as well as hematologic malignancies such as T-cell prolymphocytic leukemia (T-PLL) and non-Hodgkin lymphoma (NHL).
