ALRN-0192519 is a novel, potent, and selective small molecule allosteric inhibitor of the epidermal growth factor receptor (EGFR) specifically designed to target the L858R mutation. Developed by Allorion Therapeutics and licensed to AstraZeneca, the compound binds to a unique allosteric pocket distinct from the ATP-binding site, which allows it to inhibit EGFR signaling even in the presence of resistance mutations that render traditional tyrosine kinase inhibitors (TKIs) ineffective. ALRN-0192519 is intended for the treatment of advanced EGFR-mutant non-small cell lung cancer (NSCLC), where it may be used as a monotherapy or in combination with existing therapies like osimertinib (Tagrisso) to enhance clinical activity and overcome acquired resistance. The program is currently in the preclinical stage of development.
