ALRN-6924 + palbociclib is an investigational combination therapy evaluated in clinical trials for advanced solid tumors and lymphomas, especially those with wild-type TP53 and/or MDM2 amplification. - **ALRN-6924** is a stabilized, cell-permeating peptide that disrupts the interaction between the p53 tumor suppressor protein and its endogenous inhibitors, murine double minute 2 (MDM2) and murine double minute X (MDMX), thereby restoring p53-dependent tumor suppression in p53 wild-type tumors. - **Palbociclib** is a small molecule selectively inhibiting cyclin-dependent kinase 4 and 6 (CDK4/6), resulting in cell cycle arrest at the G1 phase. Preclinical evidence suggests this combination enhances anti-tumor activity and delays tumor growth over single-agent therapy. It has been studied primarily in patients whose disease is refractory to standard treatments, with MDM2-amplified or MDM2/CDK4-co-amplified tumors selected for expanded cohort study. Interim clinical data shows the combination is well-tolerated, with mostly mild non-hematological adverse events[1][3][5][7][9][4].
