ALS-008112

ALS-008112 is a small molecule nucleoside analog that acts as the active antiviral metabolite of lumicitabine (ALS-008176), an investigational prodrug. It is a potent and selective inhibitor of respiratory syncytial virus (RSV) polymerase. After entering epithelial cells in the respiratory tract, ALS-008112 is phosphorylated to its triphosphate form, which inhibits RSV replication by causing RNA chain termination through direct inhibition of the viral RNA-dependent RNA polymerase (L-P protein complex). The drug was initially developed by Alios BioPharma for the treatment of RSV infections. While it demonstrated significant antiviral activity in adult challenge studies, clinical trials in infants and children failed to show efficacy and revealed dose-related reversible neutropenia[1][5][10].
