Drug intelligence / Profile preview
Alstonia leaf alkaloids (also known as CALAS) is an investigational botanical drug developed by the Institute of Clinical Pharmacology of Xiyuan Hospital, China Academy of Chinese Medical Sciences (CACMS). It consists of active indole alkaloids (including scholaricine, 19-epischolaricine, vallesamine, and picrinine) extracted from the leaves of *Alstonia scholaris*. Formulated as an oral capsule, the drug is being developed for the treatment of respiratory diseases, including acute bronchitis, post-infectious cough, and asthma. It has demonstrated antiviral, antibacterial, anti-inflammatory, expectorant, antitussive, antiasthmatic, and immunoregulatory effects. Preclinical network pharmacology and pharmacodynamic studies suggest that its therapeutic mechanism may involve the modulation of targets such as cAMP-specific 3',5'-cyclic phosphodiesterases 4B and 4D (PDE4B and PDE4D), heme oxygenase 1 (HMOX1), and vascular endothelial growth factor receptor 2 (KDR).
