Altrakincept

Altrakincept is a recombinant human soluble interleukin-4 receptor (sIL-4R) developed as an anti-inflammatory biologic for the treatment of immune-mediated diseases such as asthma and graft-versus-host disease. It acts by binding to and inactivating naturally occurring interleukin-4 (IL-4), thereby blocking IL-4–mediated signaling without triggering cellular activation. This mechanism inhibits key proinflammatory pathways involved in asthma pathogenesis, including IgE class switching, VCAM-1 expression on endothelium, mucin production, 15-lipoxygenase activity, and Th2 lymphocyte stimulation leading to further cytokine release. Altrakincept was administered via inhalation (nebulized) and demonstrated safety and some efficacy in early clinical trials for moderate asthma but development was discontinued after phase I/II studies[2][3][4][6].
