Overview

ALTY-0501 is a low-dose (0.025%) topical ophthalmic solution of doxycycline, a tetracycline derivative, developed by Alacrity Biosciences for the treatment of dry eye syndrome associated with meibomian gland disease (MGD). While doxycycline is traditionally utilized as a systemic antibiotic, ALTY-0501 is designed to leverage the drug's non-antibiotic, anti-inflammatory properties at the ocular surface. Its primary mechanism of action involves the inhibition of matrix metalloproteinases (MMPs), specifically MMP-9, which are often elevated in the tear film of patients with dry eye and contribute to ocular surface damage and inflammation. By reducing MMP activity and the production of pro-inflammatory cytokines, ALTY-0501 aims to stabilize the tear film and improve ocular surface health. The drug was evaluated in Phase 2 clinical trials using the Controlled Adverse Environment (CAE) model to simulate stressful conditions for the ocular surface.
