ALVAC-HIV is a series of live recombinant canarypox virus-based HIV-1 vaccine candidates developed by Sanofi (formerly Aventis Pasteur). The vaccine utilizes the ALVAC vector, a highly attenuated canarypox virus that undergoes an abortive infection in human cells, allowing for the expression of HIV-1 antigens without viral replication. Various versions have been tested: vCP1521 (expressing clade E Env and clade B Gag/Pro), vCP205 (expressing clade B Env, Gag, and Pro), and vCP2438 (a clade C-specific version). ALVAC-HIV is most famous for its use in the RV144 Phase 3 trial in Thailand, where a prime-boost regimen with AIDSVAX B/E showed a 31.2% reduction in HIV acquisition. However, subsequent large-scale trials, such as the HVTN 702 (Uhambo) study in South Africa using the clade C version, were terminated early due to a lack of efficacy.
