ALVAC-HIV 1433 (also known as vCP1433) is a recombinant canarypox virus-based vaccine candidate developed by Aventis Pasteur (now Sanofi) for the treatment and prevention of HIV-1 infection. It utilizes the ALVAC viral vector system, a non-replicating avian poxvirus, to deliver genetic sequences of HIV-1 antigens into human cells. Specifically, ALVAC-HIV 1433 is engineered to express the HIV-1 Gag (LAI strain), Pol (LAI strain), and Env (MN strain) polyproteins. The vaccine is designed to stimulate both cellular and humoral immune responses, with a particular focus on inducing HIV-specific CD8+ cytotoxic T-lymphocyte (CTL) activity. In clinical studies such as the ANRS 117 trial, it was evaluated as part of a therapeutic immunization strategy in combination with Interleukin-2 (IL-2) and HIV lipopeptides (e.g., LIPO-6T) to improve viral control in patients on highly active antiretroviral therapy (HAART). Development of this specific construct was eventually discontinued following underwhelming results in early-to-mid-phase trials.
