ALVAC-HIV is a recombinant canarypox virus-based vaccine vector designed to express various HIV-1 antigens, including Gag, Pol, and Env. Developed by Sanofi Pasteur, the ALVAC platform utilizes a non-replicating avian poxvirus that can infect mammalian cells and express foreign proteins without producing infectious progeny, ensuring a high safety profile. The vaccine is intended to prime the immune system, particularly inducing CD8+ T-cell responses and facilitating humoral immunity when used in prime-boost regimens. It is most notable for its role in the RV144 Phase 3 clinical trial in Thailand, where a prime-boost combination of ALVAC-HIV (vCP1521) and the protein subunit vaccine AIDSVAX B/E demonstrated a modest but significant 31.2% reduction in HIV-1 infection rates.
