Overview

ALVAC-HIV vCP1521 is a recombinant canarypox virus-based HIV-1 vaccine candidate developed by Sanofi Pasteur. It is engineered to express the HIV-1 *env* (gp120) gene from the 92TH023 strain (subtype E) and the *gag* and *pro* genes from the LAI strain (subtype B). The vaccine is designed to induce both cellular and humoral immune responses against HIV-1. It gained significant prominence as the priming component of the RV144 clinical trial in Thailand, which was the first to demonstrate a modest reduction in the risk of HIV-1 infection in humans when used in a prime-boost regimen with the AIDSVAX B/E protein vaccine. While the RV144 trial showed a 31.2% efficacy, subsequent large-scale trials using modified versions of the ALVAC platform (such as HVTN 702) failed to demonstrate protection, leading to the discontinuation of several related programs.
