ALVAC-MART-1 is a recombinant canarypox virus (ALVAC) vaccine engineered to express the melanoma antigen recognized by T-cells 1 (MART-1). Developed using the ALVAC platform originally by Sanofi Pasteur, this viral vector is non-replicating in human cells, providing a safe vehicle for antigen delivery. The vaccine is designed to stimulate a systemic immune response, specifically activating MART-1-specific cytotoxic T lymphocytes (CTLs) to target and destroy melanoma cells. In clinical settings, it has been primarily investigated for the treatment of metastatic melanoma, often as part of a multi-modal immunotherapy regimen involving lymphodepleting conditioning, adoptive transfer of TCR-engineered T cells, and interleukin-2 (IL-2) to enhance the persistence and efficacy of the anti-tumor response.
