Overview

ALVAC MART-1 is a recombinant cancer vaccine developed by the National Cancer Institute (NCI) for the treatment of metastatic melanoma. It utilizes a plaque-purified canarypox virus (ALVAC) vector that has been genetically engineered to express the melanoma-associated antigen MART-1 (also known as Melan-A), a protein found in over 90% of melanoma tumors. As a biologic immunotherapy, the vaccine functions by delivering the MART-1 antigen to the patient's immune system to stimulate a targeted T-cell response. In clinical trials, it is frequently evaluated as a component of a complex regimen involving lymphodepleting chemotherapy, MART-1-specific T-cell receptor (TCR) engineered autologous lymphocytes, and high-dose interleukin-2 (IL-2), where the vaccine's role is to promote the expansion and anti-tumor efficacy of the adoptively transferred T cells.
