ALVAC-p53 is a recombinant vector cancer vaccine that uses a canarypox virus (ALVAC) as the delivery vector to express the wild-type human p53 tumor suppressor protein. The vaccine is designed to induce an immune response specifically targeting p53, which is commonly overexpressed or mutated in many human cancers. By presenting the p53 antigen, the vaccine aims to stimulate both T-cell and antibody-mediated immunity against tumor cells expressing abnormal p53. Clinical studies have shown that administration of ALVAC-p53 can elicit potent T-cell responses and IgG antibodies against both the viral vector and p53 antigen in patients with advanced cancers such as colorectal cancer. The approach has demonstrated immunogenicity but limited clinical efficacy so far, with ongoing research into its use alone or in combination with other therapies for various malignancies[1][3][4].
