Overview

**Alveltamig (ZG006)** is a trispecific T-cell engager antibody developed by Suzhou Zelgen Biopharmaceuticals that binds to **CD3** on T cells and two distinct epitopes on **DLL3** (delta-like ligand 3), a cell surface protein overexpressed on small cell lung cancer (SCLC) and neuroendocrine carcinoma (NEC) tumor cells. This biparatopic binding enhances T-cell redirection and tumor cell killing potency compared to bispecific T-cell engagers. It has shown promising antitumor activity, including a **60% overall response rate (ORR)** in a Chinese phase 2 trial for third-line SCLC and favorable safety with low cytokine release syndrome (CRS) incidence at the 10 mg dose. Alveltamig has received **FDA Orphan Drug Designation** for SCLC and **Breakthrough Therapy Designation** from China's NMPA for DLL3-positive advanced NEC post-platinum therapy.[1][2][7][10]
