Overview

ALX-0962 is a monovalent nanobody-based biologic drug candidate developed by Ablynx for the treatment of severe allergic asthma. It consists of a highly potent anti-immunoglobulin E (IgE) nanobody with a unique dual mode of action—binding to IgE with high affinity and displacing receptor-bound IgE—as well as a serum albumin-binding nanobody to extend plasma half-life in vivo. This design aimed to provide broader patient coverage, including those not eligible for existing IgE-inhibiting drugs, and potentially offer improved safety, faster onset, and more convenient dosing compared to current therapies. The drug was intended for subcutaneous administration and targeted respiratory diseases but was discontinued after preclinical development due to insufficient differentiation from competitors[1][2][3][6].
