ALX006 is a novel, symmetric 2+2 IgG1 (Fc-null) bispecific antibody designed to simultaneously target Programmed Cell Death Protein 1 (PD-1) and Vascular Endothelial Growth Factor (VEGF). Developed by Allianthera Biopharma, the drug aims to leverage the synergistic potential of immune checkpoint blockade and anti-angiogenesis to treat advanced solid tumors. By blocking the PD-1/PD-L1 pathway, ALX006 reverses tumor-induced immunosuppression and reactivates T-cells, while its VEGF-inhibitory component normalizes tumor vasculature and modulates immune cell infiltration. The molecule is engineered with an Fc-null backbone to minimize effector-mediated toxicity and optimize its safety profile. Preclinical studies presented at AACR 2026 demonstrated that ALX006 possesses superior antitumor efficacy in xenograft and syngeneic models compared to other clinical-stage bispecifics, such as ivonescimab, along with favorable pharmacokinetic properties and developability.
