ALXN2420 + somatostatin analog

ALXN2420 + somatostatin analog is a combination therapy under investigation for the treatment of acromegaly, a disorder characterized by the overproduction of growth hormone (GH) typically resulting from a pituitary adenoma. ALXN2420 is a novel, small peptide antagonist of the growth hormone receptor (GHR), designed to inhibit GH-induced signaling and thus decrease insulin-like growth factor 1 (IGF-1) levels. Somatostatin analogs (such as octreotide) are established first-line therapies that inhibit GH secretion from the pituitary gland, but are often insufficient alone to fully normalize IGF-1. Preclinical studies demonstrate that the combination has an additive effect, further reducing IGF-1 beyond what is achieved with either monotherapy. The combination is being developed by Alexion AstraZeneca Rare Disease for use in patients with acromegaly who have an inadequate biochemical response to somatostatin analogs alone[1][2][5][7].
