ALY-801 is an investigational small interfering RNA (siRNA) therapeutic being developed for the treatment of melasma and other hyperpigmentation disorders. Originally part of the Aldena Therapeutics pipeline, it is now being advanced by Alys Pharmaceuticals following the merger of several dermatology-focused biotech companies. ALY-801 utilizes a proprietary delivery platform (D-Serna) designed to enable the stable, localized delivery of chemically modified siRNA directly into the skin without the need for lipid nanoparticles. The drug works via RNA interference (RNAi) to specifically target and silence the mRNA of tyrosinase (TYR), the rate-limiting enzyme in the melanin biosynthesis pathway. By reducing the expression of tyrosinase, ALY-801 aims to decrease excessive melanin production in affected skin areas, providing a targeted approach to managing pigmentary skin conditions.
